Are you talking about semen (the whitish liquid you shoot out, which contains sperm), or are you actually talking about sperm? Sperm makes up only a small percentage of the volume of semen.

Sperm banks, I'm told, ask guys not to ejaculate for three days beforehand, so that gives a ballpark figure.

theres not much telling of how long it takes to build up to the maximum again. i think when that happens you have a wet dream, but i'm not sure. i havent had one of those since i was 13.
i once didnt masturbate for multiple weeks, and it was unbelievable when i finally did. it built up a lot. so it just gets better with each day you hold it in.

It seems with my personal observations, anyway, it takes about 2 to 3 days of not ejaculating to build a full volume ejaculation, assuming you completely depleted your semen reserves before, which in my case takes about 8 or 9 ejaculations in a day for that to happen. However, I noticed semen volume continues to increase gradually if I abstain for up to 2 weeks, then the semen volume seems to max out at about 2 weeks, and if I abstain longer than that, I don't really notice much more of an increase in volume. But the 2 week volume is much larger than a 3 day ejaculation volume. The body has the ability to vary seminal fluid production to some extent (but not sperm production - that is pretty much constant), based on demand (i.e. how often you ejaculate), and it has the ability to reabsorb excess semen and sperm that is not ejaculated.
