Not certainly - it's possible to harbour a virus but have no symptoms.

He was tested last year and is clean, and has only been with one girl since. He's had no symptons and neither has she.

Then I wouldn't worry. Of course you can't be sure without a test, but if you say he has only been with one girl after his last test, and that girl doesn't appear to have any STD, I don't see a direct reason to worry.But yeah, sleeping with one person is all it takes, so you can take a test if you worry, I'm sure he won't mind taking one if you ask.

You could suggest that you both get tested, that's a good idea at the start of any new relationship. As ineligible said, it is entirely possible for somebody to harbour a virus without any obvious symptoms. In fact there is one SDI, it may be Chlymidia, which rarely shows any symptoms at all and usually only ever becomes apparent upon testing. It's a very serious one in that, in left untreated, it can lead to infertility.
