I worry that it could just cause a layer inside the rectum which doesn't go away anymore. The waterbased lubes also work relatively well but I read that glycine isn't good for anal use cause of bacteria which could grow because of the glycine. Now I don't know what to do. Should I now rather switch to silicone?

I think you mean glycerine (which is a different substance from glycine). I don't think it should be a problem. Bacteria can grow in just about everything, but the sky doesn't fall in. The human body expects bacteria and usually handles them OK.
